Postdoctoral Research Position on new models for intracellular amyloid β in Alzheimer’s disease pathogenesis

The VIB Center for Brain & Disease Research (KU Leuven, Belgium), headed by Prof. Patrik Verstreken, is a vibrant and international research centre where key issues relevant for major human neurodegenerative diseases are tackled. The laboratory for Membrane Trafficking (Prof. Wim Annaert) studies the cellular biology of Alzheimer’s disease (AD) and aims to understand how intracellular transport regulation contributes to intramembrane proteolysis of APP and the production of neurotoxic Aβ peptides. Recent work from the lab (Sannerud et al., Cell, 2016) showed that PSEN2/γ-secretase generates the prominent pool of intracellular Aβ and familial AD-associated mutations strongly increase the accumulation of longer Aβ. To explore the pathological significance of these findings for AD pathogenesis, this project will focus on the development of novel cellular and in vivo mouse models based on familial AD mutations in PSEN2. We approach this using CRISPR/Cas9 to generate genome edited human ESCs/IPSCs which will be differentiated to long-term neuronal cultures and knock-in mouse models that will be crossed with existing APP KI models. The project encompasses a broad range of analyses, from basic neuro- and lysosomal biology to brain pathology and behavior.

We seek a young and ambitious postdoctoral fellow with experience in ESC/IPSC-derived neuronal differentiation and/or generation of knock-in mouse models. The ability to work in a multidisciplinary research team as well as effective communication skills and a good mastery of the English language are essential for a successful selection.
