Science
The study of biological systems relies to a large extent on DNA cloning technologies enabling the analysis of recombinant genes through transgenic research. Several cloning systems are available for the transfer of segments between double-stranded DNA molecules that bypass the multistep protocols involving restriction enzyme and ligase reactions. In this context, the advent of recombinational cloning methods was a significant progress because DNA fragments can now be assembled regardless of their sequence. In particular, the Gateway system was designed to join fragments in a predefined order, orientation, and reading frame. The recent development of transformation vectors and large-scale clone resources amply demonstrate that plant researchers have adopted the Gateway platform and that it will remain an important asset in projects requiring systematic cloning, modular assembly, and expression in various contexts. Within the Functional Genomics group, we continuously develop vectors based on the Gateway (TM) recombinational cloning system and designed for gene expression in plant cells. Our collection includes destinations vectors for functional gene analysis, protein fusion, silencing and promoter analysis. Recently, we have been developed a complete set of destination vectors for the analysis of monocotyledons genome, a collection of universal pEntry clones in diverse pDonor vector backbone compatible with the MultiSite Gateway system. All these resources are available to the scientific community and fully documented on line (http://www.psb.ugent.be/gateway).
These web pages also provide recombinational cloning instructions, as well as experimentally verified sequences, maps and Vector NTI files for each plasmid.
